 比对(BeRight) 2×SYBR Green PCR试剂盒 产品概述比对(BeRight) 2×SYBR Green PCR Master Mix 是采用 SYBR Green I  嵌合荧光法进行 Real Time PCR 的专用试剂。含有常温下完全封闭Taq 酶活性的热启动酶 HS Taq DNA Polymerase,能够有效抑制低温条件下引物非特异性退火或者引物二聚体引起的非特异性扩增，提高扩增反应的特异性。本试剂经过特殊配制，采用优化配方的 qPCR 专用 Buffer，大大提高了 qPCR 反应的扩增效率和检测灵敏度，可以在较宽的定量区域内得到良好的标准曲线，准确进行定量。本试剂与多数厂家的荧光定量 PCR 仪兼容，如Applied Biosystems、Eppendorf、Bio-Rad 和Roche 等。 产品组成A4004SA4004M2×SYBR Green PCR Master Mix1ml5×1ml 储存条件：本产品-20℃保存有效期至少一年，4℃可保存3个月。解冻后应充分混匀，避免产生大量气泡。 试剂原理：本产品利用热启动酶HS Taq DNA polymerase 进行 qPCR 扩增反应，通过扩增产物嵌合SYBR  Green  I 而激发的荧光信号强度进行检测。1、 PCRPCR 法是以微量 DNA 进行目的片段扩增的方法。通过 DNA 链的热变性、引物退火、DNA 聚合酶作用下引物的延伸三个步骤循环往复，可在短时间内扩增大量 DNA 片段。2、 荧光检出SYBR Green I 是一种结合与小沟中的双链 DNA 结合染料，与双链 DNA 结合后，其荧光大大增强，最大吸收波长约为 497nm，发射波长最大约为 520nm。SYBR Green I 可以与所有的双链 DNA 相结合，无需使用探针，即可实现检测，通用性好，且灵敏度很高。但是，由于 SYBR Green I 与所有的双链 DNA 相结合，因此由引物二聚体、单链二级结构以及错误的扩增产物引起的假阳性会影响定量的精确性。在定量仪器检测过程中，可以通过测量升高温度后荧光的变化，由解链曲线来分析产物的均一性，从而区分产物的溶解峰温度而区分特异与非特异的产物。 使用方法：反应条件     1、 两步法 热启动：95℃ 10  分钟；变 性：95℃ 10～20 秒；退火/延伸：60℃ 20～60 秒。溶解曲线分析。  2、 三步法热启动：95℃ 10  分钟；变 性：95℃ 10～20 秒；退 火 ：56-64℃ 10～30 秒延  伸：72℃ 10～60 秒。溶解曲线分析。对于 Roche LightCycler480，热启动时间应采用 10min，ABI7500 也可以采用 5min 热启动。qPCR反应体系配制：建议的模板量 10～100ng（基因组 DNA）或 1～10ng（cDNA 模板）。试剂20μl 体系50μl 体系终浓度2×SYBR Green PCR Master Mix10μl25μl1×Primer 1（10μM）0.4～2μl1～5μl0.2～1.0μMPrimer 2（10μM）0.4～2μl1～5μl0.2～1.0μMTemplate DNA4μl10μl-ddH2O---Total volume20μl50μl  注意事项：1、 SYBR Green PCR Master Mix 经过特殊配制，采用热启动酶，具有更高特异性；2、 对于退火温度较低的引物或超过 200bp 长片段扩增建议采用三步法； 3、 扩增前后要使用专用的区域和移液器，戴手套操作并经常更换，PCR 反应完成后切勿打开反应管。以最大限度减少 PCR 产物对样品的污染。   比对(BeRight) 2×SYBR Green PCR试剂盒
